摘要 目的研究Wnt5a在自体移植静脉重塑过程中的表达变化。方法建立大鼠颈静脉—腹主动脉移植模型,分别于术后7、14、28 d取材移植血管,采用免疫组化方法检测Wnt5a蛋白在移植血管中的表达,Real-time PCR定量检测Wnt5a mRNA的表达;以对侧颈静脉作为对照。结果免疫组化结果显示:正常静脉、术后7、14、28 d移植静脉Wnt5a阳性细胞比例分别为(4.67±0.21)%、(20.34±0.26)%、(35.25±0.20)%和(57.24±0.52)%,正常静脉与移植静脉之间差异有统计学意义(P<0.01)。Real-time PCR结果显示:术后7、14、28 d大鼠Wnt5a mRNA表达增加,分别为对照组的3.56、8.44和16.87倍。结论自体静脉移植后,Wnt5a在新生内膜及中膜中高表达,并可能与细胞增殖有关。 Objective To study the protein expression of WntSa in autogenous vein graft remodeling. Methods To establish autogenous vein graft model, 24 male SD rats of 200-250 g weight were transplanted with the jugular vein to intra renal abdominal aorta using microsur- gical technique. Graft veins were harvest at 7 d, 14 d and 28 d after transplantation. The immunohistochemistry was used to detect the WntJa expression in the vein graft. Meanwhile, WntSa mRNA was measured by quatitative Real-time PCR.The opposite normal veins served as con- trol. Results Histological staining showed that the percent of positive WntSa cells was only 4.67%±0.21%o in the normal vein,but much more in the vein graft after surgery,as 20.34%±0.26%, 35.25%±0.20% and 57.24%±0.52% ,respectively (P 〈 0.01 ). The WntSa mRNA content was also increased in vein graft to 3.56,8.44 and 16.87 folds of that in control. Conclusion WntSa is upregulated in autogenous vein grafts and may be correlated with the cell proliferation.
