补肾安胎中药对着床障碍小鼠炎性因子含量的影响

Effect of Kidney-tonifying and Miscarriage-preventing Chinese Herbal Medicine on Inflammatory Cytokine Contents in Mice with Embryo Implantation Dysfunction

目的:研究补肾安胎中药对着床障碍小鼠炎性因子含量的影响。方法:将受孕大鼠随机分为正常组、模型组、保胎灵组和坤元汤组,从妊娠第1天开始,保胎灵组和坤元汤组分别用保胎灵和坤元汤灌胃0.3mL,连续灌胃6天,正常组和模型组予0.3mL生理盐水,第3、4天小鼠注射吲哚美辛油剂复制小鼠着床障碍模型,第6天末次给药后收集小鼠血清及子宫组织,使用试剂盒检测小鼠血清及子宫组织血清干扰素-γ(IFN-γ)、白细胞介素-10(IL-10)和一氧化氮(NO)的含量。结果:与模型组比较,坤元汤组小鼠子宫重量、子宫指数及着床位点数均提高(P<0.05);坤元汤明显降低着床障碍小鼠IFN-γ和NO含量(P<0.05),显著升高IL-10含量(P<0.05)。结论:补肾安胎中药坤元汤可能通过调节炎性因子IFN-γ、IL-10及NO的含量促进胚胎着床。

Objective： To investigate the effect of kidney-tonifying and miscarriage-preventing Chinese herbal medicine onthe contents of inflammatory cytokines in mice with embryo implantation dysfunction. Methods. Conceived mice were ran- domly divided into four groups, normal group, model group, Baotailing Tablets group and Kunyuan Decoction group. From thefirst day of gestation, mice in normal and model groups revived physiological saline at 0.3 mL orally, while the mice of Bao- tailing Tablets group and Kunyuan Decoction group were orally administrated with corresponding medicine at 0.3 mL per day for6 consecutive days. On day 3 and 4, mice were injected with indomethacin oils for inducing embryo implantation dysfunction. On day 6, serum and uterus were collected after last medicine administration for the detection of contents of interferon gam-ma（IFN-γ）, intedeukin 10（IL-10） and nitric oxide（NO） with respective kits, Results： Oral administration of Kunyuan Decoc- tion significantly showed obvious effect on increasing the uterus weight, uterus index and nidation site count, reduced thecontents of IFN-γ and NO and markedly increased IL-10 content in mice with embryo implantation dysfunction （P 〈 0.05 compared with those in the model group）. Conclusion： As a kidney-tonifying and miscarriage-preventing recipe, Jybtyab Decoction could improve embryo implantation by regulating the contents of inflammatory cytokines of IFN-γ and IL-IO as well as NO.
