Establishment and Characterization of Xenograft Models of Human Neuroblastoma Bone Metastasis

摘要 目的建立模拟神经母细胞瘤(NB)骨转移的动物模型并进行鉴定。方法直接将单细胞悬浮的NB细胞系SY5Y和KCNR细胞注射到裸鼠的股骨骨髓腔内。通过放射影像学、组织学及免疫组化方法分析、评估、鉴定NB骨转移动物模型。结果 NB细胞系SY5Y和KCNR细胞导致溶骨反应,骨保护素(OPG)、RANK配体(RANKL)、甲状旁腺激素相关蛋白(PTHrP)和内皮素1(ET-1)呈强阳性表达,而CXC趋化因子受体4(CXCR4)则很少表达。结论成功建立NB骨转移动物模型,该模型可用于NB骨转移的相关研究。 Objective To develop animal models that mimic neuroblastoma (NB) bone metastasis, so as to improve the therapy of ad- vanced NB. Methods Single-cell suspensions of SY5Y and KCNR NB cells were directly injeeted into femur of nude mice. Radiologieal and histological analyses and immunohistochemistry analyses were performed to characterize these osseous-NB models. SYSY and KCNR re- sult in osteolytic responses. Results SY5Y, KCNR NB cells resulted in osteolytic reaction radiologieally and histologically. Strong expres- sions of osteopmtegerin (OPG),RANK ligand (RANKL), parathyroid hormone-related protein (PTHrP),and endothelin-1 (ET-1) were de- tected,and the expression of CXCR4 was very low in NB bone xenografts. Conclusion These animal models can be used to study NB bone metastasis biological interactions, pathways, and potential therapeutic targets, and also useful in evaluation of new agents for treatment and prevention of NB bone metastasis.
