摘要
目的对7种不同厂家壳寡糖产品(W-1~W-7)的相对分子质量、聚合度、脱乙酰度和蛋白质含量等指标进行分析检测,比较产品的质量差异,为壳寡糖质量标准的制定和产品的选择提供参考依据。方法利用高效凝胶渗透液相色谱测定相对分子质量,电喷雾质谱技术结合薄层色谱分析考察产品的聚合度分布,核磁共振波谱技术测定脱乙酰度,并对产品的蛋白含量、水分、灰分、酸碱度等进行分析检测。结果W-1、W-6及W-7的相对分子质量较小,溶解度好,寡糖聚合度分布较均匀,为较佳的寡糖产品;W-2的寡糖及低聚糖含量相当,相对分子质量分布较宽;W-3及W-4的相对分子质量较大,溶解性较差,寡糖含量低;W-5的相对分子质量虽小,但聚合度分布不均匀。结论不同厂家的壳寡糖产品在相对分子质量、聚合度和杂质含量方面各有差异,可能导致其活性及应用的差异,本研究结果将为壳寡糖的质量分析提供参考,同时为不同行业使用及选择壳寡糖提供借鉴。
Objective Compare the quality of different chitooligosaccharides (COS) by analyzing their mo lecular weight (Mr), degree of polymerization (DP), degree of deacetylation (Ddeac), and protein con centration, etc; provide data for the development of quality standards and different uses of ehitooligo saccharides. Method Determined Mr by High performance gel permeation liquid chromatography; ana lyzed DP by electrospray ionization mass spectrometry and thin-layer chromatography; measured Ddeac by nuclear magnetic resonance spectroscopy; and protein concentration, water, ash and pH were also analyzed. Result W-t, W6 and W-7 were good products with lower Mr, and uniformly distributed DP. W-2 contained both low molecular weight chitosan and COS, so the dispersion coefficient was large. The Mrs of W-3 and W-4 were high, which led to poor solubility. The Mr of W5 was low, but its I)P wasn't uniformly distributed. Conclusion COSs from different sources had different Mr, DP and impuri ties which would lead to different activities and applications. The research results are considered to pro vide references for the quality analysis and choices of COS industries.
