金银花、连翘对大肠杆菌感染模型小鼠血清Th1/Th2细胞因子的调节作用 被引量:19

摘要 目的探讨金银花、连翘对大肠杆菌感染小鼠的保护作用机制。方法将雌性昆明种小白鼠随机分为金银花组和连翘组,每组再分为正常组、中药组、模型组和治疗组4个亚组进行处理,观察小鼠的临床状态,双抗体夹心法酶联免疫吸附实验(Elisa)检测血清γ-干扰素(IFN-γ)、白细胞介素-4(IL-4)和白细胞介素-10(IL-10)的表达水平。结果金银花(0.01 kg/L)、连翘(0.01 kg/L)水煎液对小鼠大肠杆菌的感染有保护作用。与正常组及中药组比较,模型组与治疗组小鼠血清IFN-γ、IL-4表达及IFN-γ/IL-4、IFN-γ/IL-10比值均显著升高(P均<0.01);金银花治疗组较模型组IL-10表达显著升高(P<0.05),IFN-γ表达与IFN-γ/IL-4比值有升高趋势,IL-4表达和IFN-γ/IL-10比值有下降趋势;连翘治疗组较模型组IFN-γ和IL-4表达以及IFN-γ/IL-10比值有下降趋势,IL-10表达和IFN-γ/IL-4比值有升高趋势,但均无显著性差异(P>0.05)。结论感染模型小鼠辅助性T淋巴细胞1型/2型(Th1/Th2)细胞因子分泌亢进,Th1/Th2平衡有向Th1型漂移的特点。金银花、连翘水煎液均能有效保护感染模型小鼠,其调节小鼠的免疫功能可能与调节Th1/Th2平衡相关。 Objective It is to approach the protective mechanism of Flos Lonicerae and Fructus Forsythiae for mouse infected by Escherichia Coli.Methods The female Kunming mice were divided into two groups(group of Flos Lonicerae and group of Fructus Forsythiae),and each was divided into four subgroups(Normal,Medicine,Model and Cure).Then clinical status was observed and sandwich enzyme-linked immunosorbent assay(Elisa) technique was employed to detecte the expression level of Interferon-γ(IFN-γ),Interleukin-4(IL-4) and Interleukin-10(IL-10) in serum.Results The decocted fluid of Flos Lonicerae(0.01 kg/L) and Fructus Forsythiae(0.01 kg/L) had protective effects on mouse infected by Escherichia coli.Compared with Normal group and Medicine group,both expression of IFN-γ,IL-4 and ratio of IFN-γ/IL-4,IFN-γ/IL-10 were markedly rose in Model group and Cure group(all P0.01).Compared with model group,the expression of IL-10 was increased markedly(P0.05),and the expression of IFN-γ and ratio of IFN-γ/IL-4 had a upward trend,and the IL-4 expression of and ratio of IFN-γ/IL-10 had a downward trend in Flos Lonicerae Cure group;the expression of IFN-γ,IL-4 and ratio of IFN-γ/IL-10 had a downward trend,and the expression of IL-10 and ratio of IFN-γ/IL-4 were ascended in Fructus Forsythiae Cure group.There was no significant difference on them(P0.05).Conclusion Secretion of Th1/Th2 cytokines in mice of Model group is increased,and balance of Th1/Th2 is feathers drifted to Th1.The decocted fluid of Flos Lonicerae and Fructus Forsythiae can effectively protect infected model mice,the action of regulation in immune function of mice is relevant to balance of Th1/Th2.
