复方山菊对自发性高血压大鼠心脏的保护作用 被引量:1

摘要 目的:观察复方山菊对自发性高血压大鼠心脏的保护作用并分析其可能机制。方法:自发性高血压大鼠35只,随机分为SHR组(蒸馏水5 ml/kg),复方山菊高、中、低剂量组(0.45、0.15、0.05 g/kg)和卡托普利组(0.03 g/kg),每组7只。另设WKY正常对照组(蒸馏水5 ml/kg),7只。每日灌胃给药1次,连续16周。16周后,HE染色观察心肌病理变化;MASSON染色观察心肌胶原纤维;免疫组化法观察心肌转化生长因子β1(TGF-β1)、即早基因(c-fos)和诱导型一氧化氮合酶(iNOS)表达。结果:随着复方山菊剂量的增加,其心肌纤维排列逐渐整齐,结构清晰,炎性细胞浸润减少;心肌间质和血管旁的蓝色深染逐渐减少,胶原沉积现象逐渐减轻;心肌TGF-β1、c-fos和iNOS阳性细胞数表达逐渐减少。结论:复方山菊可明显减轻SHR心肌损伤,减少心肌胶原纤维的沉积,下调TGF-β1、c-fos和iNOS蛋白的表达,对心脏具有一定的保护作用。 Objective : To observe the protective effect of compound shanju( Chrysanthemi indici) on the heart injury in rats with spontaneous hypertension (SHR)and analyze the potential protective mechanisms. Methods:Thirty-five SHRs were randomized into groups of SHR( distilled water,5 ml/kg) , dosage of compound shang^u of high, medium and low (0.45,0.15,0.05 g/kg) and captopril administration (0.03 g/kg) (n =7 for each group). Another 7 wistar-kyoto(WKY) rats were included as normal controls ( distilled water, 5 ml/kg ), The animals were intragastrieally administrated with compound shangju once a day for consecutive 16 weeks. After 16 weeks,the pathological changes for myocardium were detected with HE staining. Masson's triehrome stain was used to examine the changes of myocardial collagen fibres, and immunohistoehemical technique for detecting the expression of myocardial TGF-β1, c-los and iNOS protein. Results : With increased dosage of compound shanju, neater arrangement of the myocardial fibers was observed, with clearer structure and reduced inflammatory infiltration into myocardial cells. Deeply blue staining was seen in myocardial interstitial and perivascular samples ,with reduced fibrosis and deposition of myocardial collagen. Still, the positive cell counts of TGF-β1 ,c-fos and iNOS were gradually decreased in myoeardium. Conclusion : Compound shanju can significantly reduce the incidence of myocardial injury and inhibit the myocardial collagen synthesis through down-regulation of the TGF-β1, c-fos and iNOS protein expression, suggesting that it has certain protective effects on the myocardium of rats with spontaneous hypertension.
