摘要
目的观察透骨草提取物对人肝癌SMMC-7721细胞凋亡的影响,探讨透骨草提取物诱导SMMC-7721细胞凋亡的分子机制。方法荧光显微镜观察透骨草提取物对SMMC-7721细胞形态的影响;免疫组化法检测透骨草提取物对SMMC-7721细胞Bcl-2和Bax蛋白表达的影响。结果荧光显微镜可见,40.91μg/mL透骨草提取物处理的SMMC-7721细胞,细胞皱缩,细胞核固缩,呈新月状且边集,呈现凋亡现象。免疫组化结果显示40.91μg/mL透骨草提取物处理的SMMC-7721细胞Bcl-2蛋白表达明显低于对照组(P<0.05),而Bax蛋白表达明显高于对照组(P<0.05)。结论透骨草提取物可能通过下调Bcl-2蛋白表达、上调Bax蛋白表达诱导SMMC-7721细胞凋亡。
Objective To observe apoptosis of human hepatocellular carcinoma SMMC-7721 cells induced by extractive of Phryma Leptostachya L.var.asiatica Hara and to investigate the related molecular mechanism.Methods The morphology of SMMC-7721 cells was observed with fluorescence microscope.Immunohistochemistry technique was used to determine the expressions of Bcl-2 and Bax proteins in SMMC-7721 cells.Results The fluorescence microscope showed that cell shrinkage,karyopycnosis aggregated beside edge of cell like crescent and apoptosis after SMMC-7721 cells were treated with 40.91 μg/mL extractive of Phryma Leptostachya L.var.asiatica Hara.After SMMC-7721 cells were treated with 40.91 μg/mL extractive of Phryma Leptostachya L.var.asiatica Hara,the expression of Bcl-2 protein was lower than that of control group(P〈0.05) and the expression of Bax protein was higher than that of control group(P〈0.05).Conclusion Extractive of Phryma Leptostachya L.var.asiatica Hara could induce the apoptosis of SMMC-7721 cells and the mechanism may relate to downregulating the expression of Bcl-2 protein and upregulating the expression of Bax protein.
