摘要
采用Plackett-Burman法、最陡爬坡试验和Box-Behnken设计对嗜酸乳杆菌培养基成分进行响应面优化研究。通过Plackett-Burman法筛选出影响菌体浓度的显著因素为葡萄糖、CaCO3和番茄汁。经响应面优化后,得到葡萄糖、CaCO3和番茄汁的最佳浓度分别为3.2%、0.84%和33%。优化后的增殖培养基中,菌体浓度可达4.78×109cfu/mL,是MRS配方浓度的2.18倍。
The composition of medium for Lactobacillus acidophilus was studied by Plackett-Burman method, steepest ascent experiment and Box-Behnken design. The significant factors influencing the bacterial concentration, which screened by Plackett-Burman method, were glucose, CaCO 3 and tomato juice. The optimal concentrations of these significant factors were obtained by response surface analysis as follow: glucose 3.2%, CaCO 3 0.84% and tomato juice 33%. Bacterial concentration reached 4.78×10 9 cfu/mL by using this optimal medium, which was as 2.18 times as that of MRS medium.
