复方附苓丸水提物诱导人卵巢癌SKOV3细胞凋亡作用的研究

Study on the Apoptosis of Human Ovarian Cancer SKOV3 Cell Induced by the Aqueous Extract of Compound Formula Fuling Pellet

目的:观察复方附苓丸水提物对卵巢癌SKOV3细胞的凋亡作用。方法:以未加药组为阴性对照组,用MTT法测算不同浓度药物作用于SKOV3细胞的抑制率;Annexin V/PI双染色流式细胞术检测SKOV3细胞的凋亡率;RT-PCR法测定抑凋亡基因Bcl-2 mRNA的表达。结果:复方附苓丸水提物对人卵巢癌SKOV3细胞有明显抑制作用,能明显下调Bcl-2基因mRNA表达。结论:复方附苓丸抑制SKOV3细胞生长并能诱导其凋亡,其机制与下调Bcl-2抑凋亡基因相关。

Objective：To detect the apoptosis effect of aqueous extract of compound formula Fuling Pellet on human ovarian cancer SK0V3 cell.Methods：The negative control group did not use the medicine,while the experimental group added aqueous extract of compound formula Fuling Pellet.Then MTT method was applied on measuring the inhibition ratio of medical effect on SKOV3 cell;apoptosis rate of SKOV3 cell was detected through Annexin V/PI double staining flow cytometry;mRNA expression of inhibitory apoptosis gene Bcl -2 was measured through RT-PCR method.Results：Aqueous extract of compound formula Fuling Pellet has an significant inhibitory action on human ovarian cancer SKOV3 cell,as well as downreguiating mRNA expression of Bcl-2 gene.Conclusion：Compound formula Fuling Pellet has an significant effect on inhibiting SKOV3 cell growth and inducing cell apoptosis.The mechanism may be through downreguiating inhibitory apoptosis Bcl-2 gene.
