摘要
目的:探讨海桐皮汤熏蒸对实验性兔膝关节骨性关节炎软骨细胞凋亡的影响。方法:取45只新西兰大白兔,随机取10只作为正常对照组(A组),其余35只采用Hulth方法造模,1周后取30只元膝关节感染实验兔,随机分为3组(每组10只):并随机分为模型组(B组)、水蒸气熏蒸组(C组)和海桐皮汤熏蒸组(D组)。分别采取相应的处理方法4周后处死取材,采用流式细胞术检测软骨细胞凋亡状况。结果:正常组兔关节软骨细胞凋亡率为18.57%,模型组29.41%,水治疗组和药物治疗组分别为28.26%、21.09%。与正常组相比,模型组、水蒸气熏蒸组、海桐皮汤熏蒸组有显著差异(P<0.05);与模型组相比水蒸气熏蒸组、海桐皮汤熏蒸组有显著差异(P<0.05);与水蒸气熏蒸组相比海桐皮汤熏蒸组有显著差异(P<0.05)。结论:海桐皮汤熏蒸可显著减少实验性兔膝关节骨性关节炎软骨细胞凋亡,从而延缓关节软骨的退变,促进软骨修复的作用。
Objective:To investigate the effect of Chinese compound prescription Haitongpi Decoction on apoptosis of cartilage cells in experimental rabbit knee osteoarthritis.Methods:Among forty -five New Zealand rabbits,ten were chosen randomly as control group.Another thirty-five were established knee osteoarthritis model using modified Hulth method. One week later,thirty without infection rabbits were chosen and randomly divided into 3 groups:osteoarthritis model group,vapor treated group,and Haitongpi Decoction treated group.The apoptosis of cartilage cells was detected by Flow cytometry 4 weeks later.Results:The apoptosis rate of cartilage cells was 18.57%in control group,29.41%in osteoarthritis model group,28.26%in vapor treated group,and 21.09%in Haitongpi Decoction treated group.Compared with control group,the apoptosis rates of cartilage cells in osteoarthritis model group,vapor treated group and Haitongpi Decoction treated group had significant difference(P0.05).Compared with osteoarthritis model group,the apoptosis rates of cartilage cells in vapor treated group and Haitongpi Decoction treated group had significant difference(P0.05 ).Compared with vapor treated group,the apoptosis rate of cartilage cells in Haitongpi Decoction treated group had significant difference(P0.05).Conclusion:It was considered that Haitongpi Decoction could significantly reduce the apoptosis rate of cartilage cells,potentially delay the degeneration of joint cartilage and accelerate the cartilage repair.
