摘要
熊去氧胆酸原有合成路线是从牛、羊胆汁中提取牛胆酸作为原料 ,经多步复杂反应转化为鹅去氧胆酸 ,然后对鹅去氧胆酸的羧基进行甲酯化保护 ,再经 Benkeser反应加氢还原得到熊去氧胆酸甲酯 ,最后再进行脱甲酯处理 ,得到熊去氧胆酸 ,合成的路线、工艺复杂、产率低 ;新的合成路线是由鸡胆汁中直接提取鹅去氧胆酸 ,然后选用高专一性复合型催化剂 ,加氢还原合成熊去氧胆酸 ,整个合成过程不仅大大简化合成路线和工艺、降低了成本 ,而且使熊去氧胆酸的综合转化率由原合成路线的 5 0左右提高到 65以上 .
The previous process of synthesizing ursodeoxycholic acid is very complex and the yield is very low.First,cholic acid is extracted from bile of cattle and sheep and converted to chenodeoxycholic acid through several complex reactions.Second,the carboxylic group is protected by methyl esterazation and then the protected chenodeoxycholic acid is converted to methyl ursodeoxycholate by hydrogenization with benkeser reaction.Last,ursodeoxycholic acid is obtained by demethyl ester treatment.The new synthetic process is that chenodeoxycholic acid is directly extracted and separated from bile of chicken and is reduced to ursodeoxycholic acid by hydrogenizing with a high stereospecific catalyst.The synthetic process is not only greatly simplified and cost is reduced,but the comprehensive conversion rate is increased from previous 50 percent to now over 65 percent.
