摘要
为研究淫羊藿多糖脂质体(EPSL)增强免疫的作用机理,采用MTT法和实时荧光定量PCR法测定了EPSL对鸡外周血淋巴细胞增殖及IL-2、IL-4和IFN-γmRNA表达的影响,以淫羊藿多糖(EPS)作为对照。结果表明,EPSL在体外可以显著促进淋巴细胞的增殖,其中在31.250和3.906μg·mL-1时,无论是单独作用还是协同PHA作用效果都显著优于EPS;EPSL可提高淋巴细胞IL-2、IL-4和IFN-γmRNA的表达,其中在62.500和31.250μg·mL-1时,EPSL对3种细胞因子的诱生作用显著强于EPS,EPSL在62.500μg·mL-1时的作用最强,在31.250μg·mL-1时次之,并与剂量呈正相关。EPS经过脂质体包封后其提高淋巴细胞增殖及细胞因子IL-2、IL-4和IFN-γmRNA表达的作用明显增强。
In order to study the action mechanism of epimedium polysaccharide liposome (EPSL) on immunological enhancement activity, the effects of EPSL on lymphocyte proliferation and mR- NA expression of IL-2, IL-4 and IFN-7 were determined by MTT method and real-time fluores- cent quantitation PCR assay, taking epimedium polysaccharide (EPS) as control. The results showed that EPSL could significantly enhance lymphocyte proliferation, and at 31. 250 μg ·mL-1 and 3. 906 μg · mL-1 , EPSL singly or synergistically with PHA could stimulate the lymphocyte prolif- eration which was better than EPS. And EPSL also could increase mRNA expression of IL-2, IL-4 and IFN-r, and were significantly better than EPS at 62. 500 and 31. 250 μg · mL-1. At 62. 500/μg · mL-1 , the effect of EPSL was the strongest, at 31. 250 μg·mL-1 being stronger and in a dose-depend- ant manner.
