摘要
Tetherin是新发现的具有限制多种病毒从细胞膜释放功能的细胞蛋白。本研究采用RT-PCR技术从恒河猴巨噬细胞中扩增Tetherin基因,将其插入真核表达载体pEF4/myc-His B中构建重组质粒pHF-Tetherin。并将其转染293T细胞,通过western blot和激光共聚焦分析Tetherin在293T细胞中的表达及亚细胞定位。将pHF-Tetherin分别与马传染性贫血病毒(EIAV)或猴免疫缺陷病毒(SIV)假病毒粒子感染性质粒共转染293T细胞,分析Tetherin对EIAV及SIV出芽的限制作用。研究结果表明,pHF-Tetherin转染293T细胞48 h后可以检测到Tetherin的表达并定位于细胞膜,而且该蛋白具有抑制EIAV及SIV从转染细胞中出芽的生物学功能。
Tetherin is an newly identified cellular protein which possesses a inhibition for retroviruse release from cell. To identify the biologic fimction of tetherin, tetherin gene was amplified by RT-PCR, and inserted into eukaryotic expression vector pEF4/myc-His B to construct recombinant plasmid of pHF-Tetherin, tetherin was expressed and localized on the cell membrane by confocal microscopy observation in the pHF-Tetherin transfected 293T cell. Furthermore, pHF-Tetherin were co-transfected with infectious clone of pFD-GP2 for equine infectious anemia virus (EIV) vrius-like particles (VLPs) or pBR293EAEAN for Simian immunodeficiency virus (SIV) VLPs into 293T cell, the results showed tetherin protein was able to significantly block the release of EIAV VLPs or SIV VLPs on cell surface.
