摘要
探索从豆类丝核菌培养液中分离苦马豆素的有效方法。豆类丝核菌经改良的Czapek's培养液培养14 d后,所得培养液经浓缩、超声波-水提法、三氯甲烷萃取及二氯甲烷连续液相萃取(60~70 h)等一系列操作后,得到淡黄色粗品,粗品在90℃下减压升华得到白色晶体5 mg。经薄层色谱法(TLC)和气相色谱法(GC)检测其含有苦马豆素。确定使用连续液相萃取法可以从豆类丝核菌培养液中提取分离出苦马豆素。
This experiment is conducted to find an effective method for the extraction of swainsonine from Rhizoctonia leguminicola culture. R. leguminicola were cultured on Czapek's medium, and the culture supernathant was collected after 14d. A kind of straw yellow crudes was obtained using the ultrasonic-water extraction, chloroform extraction and continuous liquid/liquid extraction procedure by dichloromethane for 60-70h. Then 5mg of white crystals was further Obtained by decompression at 90℃. The existence of swainsonine was demonstrated by thin layer ehrmnatography (TLC) and gas chromatography (GC). It suggests that swainsonine can be extracted from R. leguminicola culture by the continuous liquid/liquid extraction procedure.
