摘要 利用液体培养基摇瓶培养犁头霉属真菌 ,对培养条件进行优化 ,发现培养温度、时间以及pH值等因素对壳聚糖产量产生明显影响 结果表明 :pH值影响壳聚糖的脱乙酰程度 ,pH值控制在 4 5可获得 90 %以上的脱乙酰度 ;控制 2 8~ 2 9℃的培养温度和pH值 5 0 ,可使壳聚糖的收率达到 780mg/L 发酵法生产的壳聚糖成膜后 。 Fungus strain of A.butleri was used to produce chitosan. The culture medium contained glucose,polypepton,yeast extract and inorganic salts.The fungus strain was cultured with a reciprocal shaking at 225 r/min.Fungal mycelia was then harvested and the productivity of chitosan determined.It was found that the culturing temperature and time as well as pH of the medium all affected the productivity and qualities of chitosan.Experimental results showed that:Ⅰ.pH value influenced the degree of deacrylation of chitosan product.At pH 4.5,the yield of deacrylation of chitosan was above 90%.Ⅱ.Temperature and pH value affected the yield of chitosan.At 28~29℃, pH 5.0, the yield of chitosan was 780 mg/L. Scanning electron microscopy analysis of chitosan prepared as above showed significant difference from that prepared by extraction of shrimp. [
