摘要 目的探讨关节软骨创伤后早期(立即)行关节腔内注射透明质酸对关节软骨创伤后软骨细胞凋亡的影响。方法用2mm钻头钻孔造成兔双侧膝关节软骨急性创伤,术后立即随机选择一侧关节腔内注射1%透明质酸钠1mL(HA组),另一侧不注射任何药物(对照组)。利用末端脱氧核苷酸转移酶介导的dUTP原位切口末端标记法、流式细胞仪两种方法对受损软骨进行检测。结果治疗组软骨细胞凋亡率较对照组明显小。结论关节软骨创伤后早期(立即)进行关节腔内注射透明质酸,能有效的抑制软骨细胞的凋亡,对预防创伤后骨关节炎的发生有重要意义。 Objective To elucidate the effects hyaluronate acid(HA) on chondrocyte apoptosis following cartilage injury.Methods Adult new zealand white rabbits underwent 2 mm osteochondral drilling of the medial and lateral femoral condyles of both hind limbs.Selected one knee randomlyas experimental side,the another one as control side.The knee in experimental group was injected with 1% HA at the end of the operation.At days2,4,and2,4,12 weeks following injury apoptosis was measured with fluorescence-activated cell sorter(FACS) and the technique of Terminal-transferase dUTP Nick End Labelling(TUNEL).Results The results from flow cytometry and TUNEL assay showed that apoptosis of chondrocytes was effectively prevented by IAHA at each time point,and maximally at day 4 following injury.Conclusion Early-stage HA treatment can contribute to the prevention of chondrocytes from apoptosis and suggests a role for intra articular HA would be very important to prevent the knee from post-traumatic arthritis.
