摘要 目的研究辛伐他汀在体外对载脂蛋白A5(ApoA5)表达的调节,从而更好地理解辛伐他汀对血脂的影响。方法以含不同浓度辛伐他汀(1、5、10、25、50μmol/L)的培养液体外培养人肝癌细胞株G2(HepG2)细胞48h,并以0μmol/L辛伐他汀作为对照组,提取各组细胞总RNA和蛋白质,分别采用聚合酶链反应(PCR)和蛋白质印迹法(Western Blot)检测ApoA5的mR-NA和蛋白的表达。结果随着辛伐他汀浓度的不断增加,ApoA5的表达也不断增强(P<0.05),直至辛伐他汀浓度达到25μmol/L时,ApoA5的表达达到最大限度,再增加辛伐他汀的浓度时(50μmol/L),ApoA5表达不再增强,反而较前一浓度(25μmol/L)时的表达有所下降(P<0.05),表明辛伐他汀呈剂量依赖性上调ApoA5基因和蛋白的表达,但存在着一个最高上限浓度。结论辛伐他汀上调ApoA5基因和蛋白的表达。 Objective To investigate the regulation of simvastatin on the expression of ApoA5 m vitro,and oetecr ine mnuence of simvastatin on lipids. Methods HEPG2 cells were cultured with solution containing simvastatin of different concentrations(1, 5, 10,25,50 μmol/L) for 48 hours, with 0 μmol/L simvastatin as blank control group. Total RNA and protein from each group of cells were collected. PCR and Western Blot were used to detect the expression of apolipoprotein A5 mRNA and protein. Results The expression of ApoA5 increased along with the increase of the concentration of simvastatin (1,5,10,25,50 μmol/L) until the concentration reached 25 μmol / L,when the expression of ApoA5 was the maximum. Then as the concentration of simvastatin continued to increase (50 μmol/L),there was no more increase in the expression of ApoAS. On the contrary, the expression of ApoA5 declined compared with the former one concentration(25 μmol/L) (P〈0.05). It proved that there was an influence of does-dependant up-regulation of simvastatin on the expression of apoa5 and protein,and there was a maximum concentration. Conclusion Simvastatin raised the expression of genes and proteins of apolipoprotein A5 in hepatic cell line.
