摘要 试验用戊二醛法耦联AP和BSA,制得完全(包被)抗原AP-BSA,通过紫外扫描鉴定耦联成功,紫外吸光光度法测定得耦联比为5:1。通过方阵滴定试验确定,最佳包被浓度为4μg/mL,一抗的最适工作浓度为1:3000,二抗的最适工作浓度为1:400;通过二因素重复试验,确定用CBS做包被缓冲液,4℃下过夜为最佳包被条件。 AP was coupled with BSA by glutaraldehyde method to prepare complete (coated) antigen AP-BSA. It was identified that the coupling was successful by ultraviolet scanning and the results of determination by ultraviolet absorption spectrophotometry showed that the coupling rate was 5:1. Through phalanx titration test, it was confirmed that the best coating concentration was 4 ~g/mL, the optimum working concentration of the first antibody was 1:3 000 and the optimum working concentration of the second antibody was 1:400. It was confirmed by two-factor repeated test that the optional coating conditions were as follows : using CBS as coating buffer and overnight below 4 ℃.
