紫外灯照射牛血清白蛋白的共振光散射光谱法测定 被引量:2

摘要 利用紫外灯照射后牛血清白蛋白(BSA)的共振光散射(RLS)强度的增强,建立了一种快速、简便测定BSA的RLS光谱法。考察了pH值、反应最佳时间等因素对RLS强度的影响。实验结果表明,在pH值为4.95,紫外灯照射时间为90min条件下,测定BSA的线性范围为0.5~10mg/L,检出限为0.33mg/L。同时发现DNA与BSA共同作用时,RLS强度增强幅度增加,测定BSA的线性范围为0.05~10mg/L,检出限为0.01mg/L。将该法应用于合成样品中BSA测定,结果较好。 The detection of bovine serum albumin(BSA) under Ultraviolet (UV) irradiation system was studied by resonance light scattering(RLS) spectra. Based on the enhanced RLS intensity of the BSA under UV irradiation, a simple and fast assay for BSA determination was developed. The influences of some experimental factors,including pH value,time value on the enhancement of the RLS intensity were studied. Under the optimal conditions, the enhanced RLS intensity was proportional to the BSA concentration in the range of 0.5-10 mg/L with the detection limit of 0.33 mg/L. And in the BSA/DNA system,which was proportional to the BSA concentration in the range of 0. 05-10 mg/L with the detection limit of 0.01 mg/L. The method has been used for the detection of BSA in synthetic samples with satisfactory results.
