麻杏石甘汤对腺病毒感染的HELF细胞因子蛋白表达影响的实验研究

Experimental Research of the Impacts of Maxing Shigan Tang on the Protein Expression of HELF Cellular Factors in Adenoviral Infection

目的探讨麻杏石甘汤含药血清对3型腺病毒感染的人胚肺成纤维细胞(HELF)之转化生长因子-β1(TGF-β1)、血小板衍生生长因子-BB(PDGF-BB)、肿瘤坏死因子-α(TNF-α)蛋白表达的影响。方法以3型腺病毒攻击体外培养的HELF,制备麻杏石甘汤含药血清,作用于该细胞,另设正常细胞组,病毒对照组,利巴韦林组,采用酶联免疫吸附试验(ELISA法)检测不同组细胞TGF-β1、PDGF-BB、TNF-α的蛋白表达,并进行比较。结果病毒对照组较正常细胞组之TGF-β1、PDGF-BB、TNF-α蛋白表达均明显增强(P<0.01),含药血清组可显著降低3型腺病毒攻击后HELF的TGF-β1、PDGF-BB、TNF-α的蛋白表达(P<0.01)。结论麻杏石甘汤可下调3型腺病毒感染后TGF-β1、PDGF-BB、TNF-α的蛋白表达,这可能是其治疗病毒性肺炎的作用机制之一。

Objective To explore the impacts of the medicated serum of maxing shigan tang on the protein expressions of transformation growth factor-β1（TGF-β1）,platelet-derived growth factor-BB（PDGF-BB）and tumor necrosis factor-α（TNF-α）in human embryo lung fibroblasts（HELF）after the infection of adenovirus type 3.Methods The adenovirus type 3 was used to attack the in vitro cultured HELF.The medicated serum of maxing shigan tang was prepared and worked on the cells.A normal cell group,a virus control group and a Ribavirin group were set up separately.The enzyme-linked immunosorbent assay（ELISA）was adopted to detect the protein expressions of TGF-β1,PDGF-BB and TNF-α in the cells of each group and the comparison was made.Results The protein expressions of TGF-β1,PDGF-BB and TNF-α in the virus control group were stronger apparently than those in the normal cell group（P〈0.01）.The protein expressions of TGF-β1,PDGF-BB and TNF-α of HELF after the attack of adenovirus type 3 were reduced significantly in the medicated serum group（P〈0.01）.Conclusion Maxing shigan tang down-regulates the protein expressions of TGF-β1,PDGF-BB and TNF-α of HELF after the infection of adenovirus type 3,which is probably one of the mechanisms of its efficacy on viral pneumonia.
