慢性移植物肾病患者外周血淋巴细胞细胞因子表达的研究 被引量:2

摘要 目的探讨慢性移植物肾病(chronic allograft nephropathy,CAN)患者外周的血淋巴细胞细胞因子的表达情况。方法选取2004年7月至2005年8月在中国人民解放军第309医院器官移植研究所就诊的15例CAN和22例移植肾功能正常的肾移植受者,分别列为CAN组和非CAN组。所有患者均签署知情同意书,符合医学伦理学规定。两组患者分别抽取空腹外周血4ml,分离外周血淋巴细胞。采用实时荧光定量聚合酶链反应比较两组患者外周血淋巴细胞细胞因子白介素(IL)-2、IL-10、干扰素(IFN)-γ及转化生长因子(TGF)-β1的信使核糖核酸(messenger RNA,mRNA)表达的差异。结果与非CAN组比较,CAN组的IL-2和IL-10 mRNA表达差异无统计学意义(均为P>0.05),IFN-γ、TGF-β1 mRNA表达明显较高,差异有统计学意义(分别为P<0.05、P<0.01)。结论 IFN-γ、TGF-β1 mRNA的高表达可能与CAN的发生、进展有关。 Objective To investigate the expression of cytokine of peripheral blood lymphocyte (PBL) in patients with chronic allograft nephropathy (CAN). Methods Thirty-seven renal transplant recipients were selected in the Institute of Organ Transplantation of the 309th Hospital of Chinese PLA from July 2004 to August 2005, who were divided into CAN group (n = 15, recipients with CAN) and non-CAN group (n =22, recipients with normal renal function). This study was approved by local ethical committee and the informed consent of all participating subjects was obtained. Fasting peripheral blood (4 ml) was drawed from patients in 2 groups and PBL was separated. The messenger ribonucleic acid (mRNA) expression of eytokines in PBL, including interleukin (IL) -2, IL-10, interferon(IFN) -γand transforming growth factor(TGF) - β1, was detected by real-time fluorescent quantitative polymerase chain reaction, and the difference between the two groups was compared. Results There was no significant difference in the expression of IL-2 mRNA and IL-10 mRNA between CAN group and non-CAN group ( all in P 〉 0. 05 ). Compared with non-CAN group, the expressions of IFN-γmRNA and TGF-β1 mRNA, were significantly higher in CAN group (P 〈 0. 05, P 〈 0. 01 ). Conclusions The high expression of IFN-γ, mRNA and TGF-β1 mRNA may have relation with the development and progression of CAN.
