摘要
目的探讨乳源免疫调节肽(PGPIPN)对人卵巢癌细胞株(SKOV3)侵袭和转移的影响。方法 Transwell小室测定其侵袭力和迁移力,细胞划痕实验测定细胞运动能力,细胞克隆形成实验观察SKOV3的克隆形成能力,用MTT法比较PGPIPN对SKOV3、人正常肝细胞株LO2和小鼠永生化成纤维细胞株(MEFS)的生长抑制情况,检测PGPIPN的细胞毒性。结果 Transwell试验中,与空白对照组相比,浓度为1 mg.L-1和10 mg.L-1的PGPIPN明显抑制了SKOV3的侵袭和转移(P<0.01),抑制率分别为20.20%和23.78%;划痕实验显示对细胞的运动能力得到了明显的抑制;平板法的克隆形成实验结果较明显,与空白对照组相比,1 mg.L-1和10 mg.L-1的PGPIPN抑制率分别为32.50%和38.11%;PGPIPN抑制SKOV3细胞生长,对正常细胞无明显影响。结论乳源免疫调节肽能够抑制人卵巢癌细胞的侵袭和转移。
Aim To investigate the effects of immunom- modulating peptide (PGPIPN) from B-casein on inva- sion and metastasis of ovarian papillary serous cystade- nocarcinoma cells (SKOV3 ). Methods The cell in- vasion and migration of SKOV3 were determined by tr- answell chamber model. The cell crawling capability was determined by cell scratch assay. The ability of colony formation was assayed by colony experiment. The cell toxicities of PGPIPN were assayed by compa-ring the inhibitory effects of PGPIPN on SKOV3, nor- mal human hepatic cell line L02 and mouse embryo fi- broblasts (MEFS) proliferations with MTY method. Results Compared with control group, PGPIPN at lmg · L-1 and 10 mg · L-1 significantly inhibited cell invasion and migration (P 〈 0.01 ) , in which the in- hibitory percentage was 20. 20% and 23.78%, respec- tively, in transwell chamber model. In cell scratch as- say , PGPIPN obviously inhibited cell movement . Incolony formation, results were very distinct, the inhibi- tory percentage Of PGPIPN at 1 mg · L-1 and 10 mg· L-1 was 32.50% and 38. 11% respectively, compared with control groups. The inhibitory effects of PGPIPN on SKOV3 cells proliferation were not observed on thegrowth of normal cells (human hepatic cell line LO2, MEFS ) by MTT assay. Conclusions PGPIPN can inhibit invasion and migration of SKOV3 cells in vitro.
