摘要
目的探讨罗格列酮对阻塞性睡眠呼吸暂停综合征(OSAS)陆川猪模型血清和脂肪组织抵抗素、瘦素mRNA及蛋白表达量的影响。方法选取23只陆川猪,随机分为:正常对照组(6只)、模型组(6只)、罗格列酮干预组(6只)。罗格列酮干预组喂服罗格列酮0.2 mg.kg-1.d-1;正常对照组及OSAS模型组给予等量生理盐水,每天1次。12周后处死各组实验动物并取材。采用多导睡眠呼吸监测仪进行监测,免疫放射分析法检测3组血清抵抗素、瘦素水平的变化,分别用RT-PCR法、Western blot法检测大网膜脂肪组织抵抗素、瘦素基因及蛋白的表达。结果与对照组比较,OS-AS猪模型血清抵抗素、瘦素水平升高,脂肪组织抵抗素、瘦素mRNA及蛋白表达灰度值升高,差异有统计学意义(P<0.01);与模型组比较,罗格列酮干预治疗后,呼吸暂停低通气指数(AHI)降低,最低血氧饱和度(MinSaO2)升高(P<0.05);血清抵抗素、瘦素水平下降、脂肪组织抵抗素mRNA、瘦素mRNA及蛋白表达灰度值降低(P<0.05),但仍高于对照组水平(P<0.05)。结论罗格列酮可能通过减轻胰岛素抵抗,下调血清和脂肪组织抵抗素、瘦素mRNA及蛋白表达,从而发挥其对OSAS的干预作用。
Aim To study the effects of rosiglitazone on the resistin, leptin mRNA and protein expression in se- rum and adipose tissue of obstructive sleep apnea syn- drome(OSAS) Lu Chuan pig model. Methods Twen- ty-three Lu Chuan swines were randomly divided into control group (six) , model group (six) and rosiglita- zone intervention group (six). The rosiglitazone treat- ment group were fed with rosiglitazone 0. 2 mg · kg-1 · d-m, and the control group and OSAS model group were fed with equal volume of normal saline once a day. Each group was sacrificed after 12 weeks and ma- terials were drawn. Using polysomnography respiratory monitor to monitor, Resistin leptin level in serum in each group were detected using immunoradiometric as- say method. Resistin, leptin gene and protein expres- sion were assayed respectively by RT-PCR and Western blot method in omental adipose tissue. Results Corn-pared with the control group, resistin, leptin levels in serum of OSAS model group were significantly in- creased. Resistin, leptin mRNA and protein expression gray value in adipose tissue were raised, and the differ- ence was statistically significant ( P 〈 0.01 ). Com- pared with OSAS model group, after rosiglitazone treat- ment, sleep apnea hypopnea index (AHI) decreased, the lowest blood oxygen desaturation (SaO2 ) increased (P 〈 0.05 ), blood resistin, leptin levels dropped, ResistinmRNA, LeptinmRNA and protein expression gray value in adipose tissue declined (P 〈 0. 05 ), but still higher than control groups ( P 〈 0. 05 ). Conclu- sion Rosiglitazone can make Resistin, leptin mRNA and protein expression reduced by alleviating insulin resistance, to play its role of intervention in OSAS.
