反复自然流产患者IL-17、T-bet、GATA-3和FoxP3mRNA表达水平的检测

Detection of IL-17,T-bet,GATA-3 and FoxP3 mRNA in patients with recurrent spontaneous abortion

目的探讨反复自然流产患者辅助性T细胞17(Th17)免疫应答情况及其与Th1、Th2和Treg细胞相互之间的关系。方法实时荧光定量PCR(Realtime PCR)检测反复自然流产患者30例(观察组)及健康早孕体检者38例(对照组)外周血中的IL-17、T-bet、GATA-3和FoxP3 mRNA表达水平。结果观察组IL-17mRNA、T-bet mRNA表达水平较对照组明显升高,差异有统计学意义(P<0.0001)。而GATA-3 mRNA、FoxP3 mRNA表达水平显著低于对照组(P<0.0001);观察组Th1/Th2和Th17/Treg的比值均较对照组显著升高,差异有统计学意义(P<0.0001)。结论 Th17细胞参与了反复自然流产的免疫应答,Th1/Th2和Th17/Treg细胞之间的平衡在妊娠的维持中发挥重要作用。

Objective To study the immune response of T-helper cell 17 （Th17） and the relationship of Thl, Th2 and Treg cells in recurrent spontaneous abortion （RSA） patients. Method qPCR was used to analyze the expression levels of IL-17, T-bet, GATA-3 and FoxP3 mRNA in 30 RSA patients and 38 healthy controls. Results The levels IL-17 and T-bet mRNA in the RSA group were significantly higher than those in the control group （P〈0.0001）. However, the mRNA levels of GATA-3 and FoxP3 in the RSA group were significantly lower than the control group （P〈0.0001）. The ratios of Thl/Th2 and Th17/Treg in the RSA group were significantly different from those in the control group （P〈 0.0001 ）. Conclusion Thl7 cells are involved in the immune response of RSA, and the balance between Thl/Th2 and Th17/Treg cells is important in RSA.
