摘要
以库伦大三棱荞麦为研究对象,以子叶、下胚轴为外植体诱导愈伤组织,为建立荞麦的遗传转化体系奠定基础。研究结果表明:去皮荞麦种子的最适消毒条件是0.1%升汞消毒1.5min;以子叶为外植体诱导愈伤组织的最适培养基为MS+30g/L蔗糖+7g/L琼脂粉+1.2mg/L6-BA+0.05mg/L NAA+0.1mg/L2,4-D;以下胚轴为外植体诱导愈伤组织的最适培养基为MS+30g/蔗糖+7g/L琼脂粉+0.5mg/L6-BA+0.05mg/LNAA+0.05mg/L2,4-D;下胚轴比子叶更适合作为外植体诱导愈伤组织。
The Kulun Big Triquetrous Buckwheats(Fagopyrum esculentum Moeneh.)is the object researched,and its cotyledons and hypocotyls treat as explants to induce the callus,the research aimed to study the conditions that affected the establishment of the heredity converts system of buckwheats.Results of the study:the best disinfection conditions is 0.1% HgCl2 1.5min on Peeled buckwheats seeds;and the optimal medium for embryogenic callus induction is MS+30g/Lsucrose+7g/Lagar+1.2mg/L6-BA+ 0.05mg/LNAA+0.1mg/L2,4-D with cotyledons for explants to induce callus;and the best medium for embryogenic callus induction is MS+30g/L sucrose+7g/L agar +0.5mg/L6-BA +0.05mg/LNAA+0.05mg/L 2,4-D with hypocotyls for explants to induce callus;hypocotyls is better than cotyledons as explant to induce callus.
