甘草的醇沉淀物体外抗RSV有效部位的筛选 被引量:6

摘要 目的筛选甘草的醇沉淀物经柱层析后具有体外抗呼吸道合胞病毒(RSV)感染作用的洗脱部分。方法采用水提醇沉法制出甘草的醇沉物,经大孔吸附树脂HPD722、HPD750、D101柱层析、蒸馏水洗脱出多个部位;以利巴韦林为阳性对照药物,采用MTT法检测洗脱部分与甘草酸在Hep-2细胞中的药物毒性及抗RSV效果。结果 HPD722水部位、HPD750水部位、D101水部位、甘草酸、利巴韦林对RSV感染的治疗指数(TI)依次为13.698、261.578、1.461、1.708、27.895。结论甘草醇沉物在体外对RSV具有显著的抑制作用,HPD750水部位可能是其主要的有效部位,值得进一步深入研究。 Objective To identify the eluted components made by column chromatography from sediments of Radix Glycytthizae and their anti-respiratory syncytial virus(RSV) activities in vitro.Methods The sediments were boiled with distilled water,precipitated with alcohol,and then eluted with distilled water through column chromatography with macroporous resins HPD722,HPD750 and D101.The toxicity and anti-RSV effect of eluted components and glycyrrhizic acid were determined in Hep-2 cells by MTT assay.Ribavirin was used as the positive control drug.Results The therapeutic indexes(TI=TC50/EC50) of HPD722 water component,HPD750 water component,D101 water component,glycyrrhizic acid and Ribavirin against RSV were 13.698,261.578,1.461,1.708 and 27.895,respectively.Conclusion The inhibition effects of Radix Glycytthizae sediments on RSV in Hep-2 cells are obvious,and HPD750 water component may be the most effective one and deserve to further study.
