摘要
为探讨创伤弧菌Vibrio vulnifiticus外膜蛋白(outer membrane protein,OMP)免疫刺激复合物(Immune stimulating Complexs,ISCOMs)的抗原性,制备创伤弧菌OMP-ISCOMs腹腔注射免疫欧洲鳗鲡Anguilla anguilla(平均规格为17g·尾-1),通过免疫血清学检测和攻毒保护试验,测定ISCOMs最小免疫剂量。结果表明,于免疫后第30d用FJ03-X2菌株攻毒,免疫组40、20、10、5μg·尾-1的免疫保护力分别为100%、87.5%、75%、50%;免疫血清效价分别为1∶6 400、1∶3 200、1∶800、1∶200。结果说明创伤弧菌外膜蛋白ISCOMs的最小免疫剂量为0.59μg.g-1水温(24±1)℃,并证实了创伤弧菌OMP-ISCOMs具强抗原性。
Antigenicity of the immune stimulating complexes in the outer membrane protein(OMP-ISCOMs) of Vibrio vulnifiticus was studied by injecting the vaccine into the European eels(Anguilla anguilla).The minimum dosage required for effective vaccination was determined by serology and immune protection test.The immune response was dosage dependent.Thirty days after the eels were challenged with the V.vulnificus FJ03-X2 strains at 200,100,50 and 25 μg·mL-1 to result in 100%,87.5%,75% and 50% of protection,respectively;and the titers of serum antibody of 1∶6400,1∶3200,1∶800 and 1∶200 in turn,respectively.It appeared that the minimum dose of the OMP-ISCOMs of Vibrio vulnifiticus vaccine was 0.59 μg·g-1.
