大剂量强的松影响小鼠MHCI类基因表达的实验研究

摘要 目的 研究大剂量糖皮质激素药物强的松对小鼠MHCI类基因表达的影响。方法25只BALB/c小鼠随机分成两组,对照组10只,实验组15只,实验组每天给予强的松(20mg!kg)灌胃,共给药30天。实验结束后取肝脏、脾脏、肾脏和肺组织少许提取RNA,RT—PCR测定MHCI类基因mRNA的表达,Western印迹测定它们的蛋白表达。结果实验结束时,肝脏、脾脏、肾脏和肺组织的MHCI类基因mRNA表达分别为0.06、0.05、0.09和0.05,明显低于对照组;MHCI类基因蛋白表达分别为9.13、11.35、9.86和9.36,明显低于对照组。结论大剂量强的松能降低小鼠机体肝脏、脾脏、肾脏和肺组织的MHCI类基因mRNA和蛋白表达。 Objective To study mouse MHC class I gene expression affected by high dose glucocorticoid prednisone. Methods Twenty five BALB/c mice were randomly divided into two groups, 10 mice served as the control group, 15 mice in experimental group. Experimental group were given daily doses of prednisone (20mg/kg) for 50 days. 30 days later the mice were killed and their livers, spleens, kidneys and lungs were taken out, total RNA was extracted, RTPCR and Western blotting were used to examine the mRNA and protein expression of MHC class I gone. Results When the experiment was end, liver, spleen, kidney and lung MHC class I gene mRNA expression lever in experimental group were 0.06,0.05,0.09 and 0.05 respectively, showed obviously lower than in control group, MHC class I gene protein expression lever in experimental group were 9.13,11.35,9.86 and 9.36 respectively, showed obviously lower than in control group. Conclusion High dose prednisone can decrease mouse organism liver, spieen, kidney and lung MHC class I gene mRNA and protein expression.
