摘要
【目的】检测外周血单个核细胞(peripheral blood mononuclear cell,PBMC)免疫球蛋白转录子4(immunoglobulin-like transcript-4,ILT-4)mRNA在肝癌患者中的表达,并探讨其在肿瘤发生发展中的作用。【方法】以β-actin为内参照,实时荧光定量RT-PCR方法检测肝癌患者和健康体检者外周血单个核细胞ILT-4mRNA的表达。【结果】正常对照组ILT-4mRNA表达水平(0.450±0.216)显著低于肝癌组(0.887±0.253,P<0.05),肝癌患者组在不同分期之间的表达差异无统计学意义(P>0.05),低分化(1.061±0.126)显著高于高、中分化(0.693±0.215,P<0.05)。【结论】肝癌患者外周血PBMCILT-4mRNA表达水平上调,检测PBMC的ILT-4基因表达水平有助于肝癌的诊断和研究。
[Objective]To investigate the expression level of ILT-4 in PBMC of liver cancer patients and its clinical significance. [Method]The mRNA of ILT-4 in PBMC were detected by real-time relative quantitative PCR method in 38 patients with liver cancer and 40 healthy individuals. [Result]The relative expression level of ILT-4 in PBMC in liver cancer group(0.887±0.253) was significantly higher than healthy group(0.450±0.216, P 0.05 ). The relative expression level of ILT-4 in PBMC in poorly differentiated group(1.061±0.126)was significantly higher than hyspo- and moderately differentiated group(0.693±0.215, P 0.05 ).[Conclusion]ILT-4 may play a critical role in the tumor immunity response. It may provide a novel diagnosis and research tool for liver cancer.
