摘要
目的探讨半枝莲提取物(SBE)诱导人大肠癌细胞系(SW480、LoVo、HT-29)的凋亡作用及其可能机制。方法体外培养SW480、LoVo、HT-29细胞,以药物康复新、露蜂房作对照,应用MTT法和流式细胞术检测不同浓度SBE处理三种细胞48h后的抑制率、细胞凋亡率及周期变化;RT-PCR检测SBE处理后凋亡相关基因Bcl-2和Caspase-3mRNA表达。结果与康复新、露蜂房比较,SBE对三种细胞的抑制率和细胞凋亡率均呈剂量依赖性显著升高(P<0.01);S期细胞含量增多,出现S期阻滞;Caspase-3 mRNA表达亦显著上升,Bcl-2表达明显下降(P<0.01)。结论SBE具有诱导大肠癌细胞系凋亡和抑制增殖作用,作用强于康复新和露蜂房提取物。其机制可能与Caspase-3基因表达的上调及Bcl-2基因表达下降有关。
Objective To explore the effects and underling mechanism of Scutellaria barbata D.Don extract(SBE) on cell apoptosis of human colon carcinoma cell lines SW480,LoVo and HT-29. Methods The cells of SW480,LoVo and HT-29 were cultured and treated by SBE,Medicine Kang Fuxin and nidus Vespae for 48 hours.The apoptosis and inhibitory rates and the change of cell cycle were evaluated by MTT assay and flow cytometry.The expressions of Bcl-2 and Caspase-3 were detected by RT-PCR. Results Compared to Medicine Kang Fuxin and nidus Vespae,the apoptosis and inhibitory rates of SW480,LoVo and HT-29 cells treated with different concentrations of SBE were increased in a dose-dependent manner(P〈0.01).The cells stayed at S stage were increased,the expression of Bcl-2 was reduced and Caspase-3 expression was increased in the SBE-treatead cells(P〈0.01).Conclusion SBE can induce apoptosis and inhibit proliferation of human colon carcinoma cell lines,which might be related to upregulating the expression of Caspase-3 and downregulating the expression of Bcl-2.The inhibitory effect of SBE is greater than that of the extracts of Medicine Kang Fuxin and nidus Vespae.
