半边莲生物碱对高血压鼠脑动脉血管重构的作用 被引量:5

摘要 目的:观察半边莲生物碱对高血压大鼠脑动脉内皮素-1(endothelin-1,ET-1)mRNA表达的影响,探讨半边莲生物碱对高血压大鼠脑动脉血管重构的作用。方法:将16只高血压模型鼠随机分为2组:半边莲生物碱组和高血压实验组,每组8只。另外,随机选取8只正常血压大鼠作为正常对照组。半边莲生物碱组给予半边莲生物碱,高血压和正常对照组大鼠组给予等量生理盐水,各组大鼠均灌胃给药,每日1次,连续给药8周。8周后,分别通过Western blot和RT-PCR技术检测各组大鼠脑基底动脉ET-1蛋白和ET-1mRNA的表达。结果:半边莲生物碱组大鼠脑基底动脉ET-1蛋白及ET-1 mRNA的表达较高血压组明显降低(P<0.05)。结论:半边莲生物碱可抑制高血压鼠脑基底动脉ET-1蛋白及ET-1 mRNA的表达,提示半边莲生物碱在预防和治疗高血压诱发的脑血管重构中有重要的作用。 Objective: To explore the effects of Lobelia Chinensis Lout Alkaloids(LCLA) on vascular remodeling of cer- ebral artery in hypertensive rats, and to observe the influence of LCLA on the expression of endothelin-1 (ET-1) mRNA of cerebral artery in hypertensive rats. Methods : Sixteen hypertensive model rats were divided into 2 groups randomly, in- cluding the LCLA group and hypertensive group( 8 for each group). Eight Wistar normotensive rats were selected as con- trol group. LCLA were administered for LCLA group, and 0.9% NaC1 solution for hypertensive group and control group. Drug was administered with stomach for every group rats, one time for each day and continuous for 8 weeks. After 8 weeks, the expressions of ET-1 and ET-lmRNA in the cerebral basilar artery for each group rats were observed using Western blot and RT-PCR respectively. Result: Compared with hypertensive group rats, ET-1 and ET-1 mRNA expres- sions in cerebral basilar artery were significantly decreased in LCLA group rats ( P 〈 0.05 ). Conclusion : The LCLA can inhibit the expression of ET-1 and ET-1 mRNA in the cerebral basilar artery in hypertensive rats, which suggests that the LCLA plays an important role in preventing and treating the cerebral vascular remodeling induced by hypertension.
