睡眠剥夺对大鼠自发活动及各脑区腺苷A_1受体表达的影响 被引量:2

摘要 目的:探讨大鼠不同脑区腺苷A1受体在睡眠调节过程中的作用。方法:SD雄性大鼠(24只)随机分为睡眠剥夺组、大平台对照组和正常对照组。采用小平台水环境法对大鼠进行72h睡眠剥夺,用大平台水环境作对照。用开场实验观察睡眠剥夺后大鼠自发活动变化,并用逆转录-聚合酶链反应法检测各组前额叶、海马、下丘脑、中脑区、纹状体等脑区腺苷A1受体mRNA浓度变化。结果:睡眠剥夺组自发活动总路程大于大平台对照组((26 100±823)mmvs(15 656±729)mm,P<0.05),自发活动中央路程及休息时间两组比无统计学意义。腺苷A1受体mRNA表达结果显示:在海马,睡眠剥夺组低于大平台对照组((0.18±0.02)vs(0.93±0.17),P<0.05),大平台对照组高于正常对照组((0.93±0.17)vs(0.26±0.05),P<0.05);在中脑,睡眠剥夺组低于大平台对照组((0.06±0.00)vs(0.16±0.02),P<0.05),大平台对照组低于正常对照组((0.16±0.02)vs(0.26±0.03),P<0.05);在额叶,睡眠剥夺组与大平台对照组比差别无统计学意义,大平台对照组低于正常对照组((1.74±0.20)vs(2.68±0.39),P<0.05);在下丘脑和纹状体,各组差别无统计学意义。结论:睡眠剥夺对大鼠各脑区腺苷A1受体表达的影响不同,海马和中脑腺苷A1受体可能参与了睡眠调节。大鼠睡眠剥夺后的自发活动改变与腺苷A1受体表达改变的关系需进一步研究。 Objective:To investigate the influence of adenosine A1 receptor on adjusting sleep.Methods:Twenty-four SD rats were randomly divided into sleep deprivation group,rank-plat control group and normal control group.The small platform method was used to establish sleep deprivation model.The open field test was applied to evaluate the behavior of the model rats.The expression level of adenosine A1 receptor mRNA was studied by reverse transcription polymerase chain reaction.Results:The total distance of sleep deprivation group compared with rank-plat control group increased((26 100±823)mm vs(15 656±729)mm,P0.05).The rest time and the central distance had no differences.The adenosine A1 receptor expression level resulted that the sleep deprivation group was less than rank-plat control group((0.18±0.02)vs(0.93±0.17),P0.05)but the rank-plat control group was higher than the normal group((0.93±0.17)vs(0.26±0.05),P0.05) in the hippocampus,the sleep deprivation group was less than rank-plat control group((0.06±0.00)vs(0.16±0.02),P0.05)and rank-plat control group was less than the normal group((0.16±0.02)vs(0.26±0.03),P0.05)in the midbrain,there was no difference between sleep deprivation group and rank-plat control group but rank-plat control group was less than the normal group((1.74±0.20)vs(2.68±0.39),P0.05)in the prefrontal cortex,there was no difference among groups in hypothalamus and striate.Conclusion:After sleep deprivation,the adenosine A1 receptor in different part of rat brain has different change.The adenosine A1 receptor in the hippocampus and midbrain may take part in sleep regulation.The relationship between the changes of adenosine A1 receptor and the behavior after sleep deprivation should be further investigated.
