摘要
目的研究人婆罗双树样基因(SALL4)mRNA及其蛋白在儿童急性白血病(AL)中的表达情况。方法运用逆转录聚合酶链式反应(RT-PCR)技术对急性白血病患儿及正常对照的单核细胞进行SALL4 mRNA表达测定,运用ELISA法同步检测患儿血清中SALL4蛋白表达水平,并对其相关性进行分析。结果①AL组与对照组之间SALL4 mRNA表达率比较差异有统计学意义(P〈0.05),其中急性淋巴细胞白血病组(ALL)和急性非淋巴细胞白血病(ANLL)组均明显高于正常对照组(P〈0.05),B—ALL和T—ALL之间的SALL4 mRNA表达率比较差异有统计学意义(P〈0.05),而B—ALL与ANLL组间比较差异无统计学意义(P〉0.05)。②B.ALL组、T-ALL组、AN—LL组、正常对照组的SALL4蛋白表达量均数分别为:136±17、33±23、151±64、55±16。③各组中的SALL4 mRNA和外周血清中的SALL4蛋白表达呈正相关(r=0.931,P〈0.01)。结论儿童AL骨髓单个核细胞中存在SALIA基因的异常高表达,与健康对照比较差异有统计学意义,提示SALL4基因参与了儿童AL的发病。儿童AL中,SALL4mRNA及其蛋白在B—ALL和ANLL中高表达,且呈正相关;而在T—ALL和健康对照中不表达,提示儿童AL中B—ALL和T—ALL的发病机制可能不同;而B—ALL和ANLL可能存在同样的发病机制。
Objective To study the mRNA and protein expression of sal-like gene 4 (SALL4) in children's acute leukemia (AL). Methods Through the real-time semi-quantitative reserve transcriptase-polymerase chain reaction(RT-PCR) method to test the mRNA expression level of SALL4 in the bone marrow of children' s AL and normal control group, explore the protein expression level of SALL4 in enzyme-linked immuno sorbent assay(ELISA), and proceed the correlation analysis. Results (1)There was a statistically significant expression between AL and control group ( P 〈 0.05 ), the mRNA expression levels of SALL4 in ALL and ANLL were significantly higher than those in control group ( P 〈 0. 05 ), there was a statistically significant expression between B-ALL and T-ALL ( P 〈 0. 05 ), but the mRNA expression level of SALIA has no difference between B-ALL and ANLL ( P 〉 0. 05 ). (2)The amount of SALL4 protein expression in B-ALL, T-ALL, ANLL was respectively 136 ± 17, 33 ±23, 151 ±64, 55 ± 16. (3) The mRNA and protein expression of SALL4 was positively correlated( r =0. 931, P 〈0.01 ). Conclusions There is abnormal higher expression of SALL4 in children' s AL bone marrow mononuclear ceils, it is statistically significant compared with the normal control group, and suggest that SALIA is involved in the pathogenesis of children' s AL. In children' s AL, the mRNA and protein expression of SALL4 is higher in B-ALL and ANLL, and positively related, but they don't express in T-ALL and the normal control group. It suggests that the pathogenesis in children' s B-ALL and T-ALL may be different, and the pathogenesis in children' s B-ALL and ANLL may be same.
