Objective To evaluate the safety of celecoxib on gastrointestinal tract in aged rats.Methods Forty aged SD rats were assigned in to 4 groups(n=10 in each group): the rats were fed by gavage with NS,2% celecoxib,0.1% indomethacin or 10% cimetidine,respectively.In cimetidine group the animals were also fed with 0.1% indomethacin 1h after feed with cimetidine.The changes of gastric mucosa were examined by light and scan electron microscopy,and the lesion index was assessed.The gastric mucosal blood flow(GMBF),the 6-keto-Prostaglandin F1α(6-keto-PGF1α) and thromboxane B2(TXB2) levels in gastric mucosa were also measured.Results The injury of gastric mucosa in indomethacin group was more severe than that in the celecoxib group.Compared to celecoxib group,the GMBF,the 6-keto-PGF1α and TXB2 levels the indomethacin and cimetidine groups were significantly decreased(P0.01 or P0.05).Comparing to control group,the GMBF in celecoxib group was significantly decreased(P0.01),but the 6-keto-PGF1 and TXB2 levels in gastric mucosa were not changed significantly(P0.05).Conclusion COX-2 inhibitor celecoxib induces less damage for gastric mucosa compared to traditional nonsteroidal anti-inflammatory drugs(NSAIDs).
