摘要
目的分别比较热休克蛋白60(HSP60)在多西紫杉醇(Docetaxel)诱导前列腺癌PC-3细胞凋亡及耐药前后表达的差异,并探讨其意义。方法采用20μmol/L Docetaxel诱导PC-3细胞凋亡,后采用逐渐加量的方式培养PC-3细胞Docetaxel耐药株;采用双向荧光差异凝胶电泳(DIGE)技术定量筛选未处理组PC-3细胞与凋亡PC-3细胞,及PC-3细胞敏感株与耐药株的差异蛋白,并采用基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)技术对其进行成分鉴定,进而检测HSP60的表达。结果 HSP60在Docetaxel诱导PC-3凋亡细胞中表达上调,在Docetaxel耐药株中表达下调。结论 HSP60在PC-3凋亡细胞中高表达,在Docetaxel耐药株中的低表达,提示其可能与PC-3细胞的凋亡及耐药有重要关系。
Objective To identify the different expressions of heat shock protein 60(HSP60) in apoptosis and drug resistance induced by docetaxel in androgen independent prostate cancer PC-3 cells,and investigate its significance.Methods Apoptotic PC-3 cells were induced by 20 μmol/L docetaxel.Docetaxel-resistant PC-3 cells were produced by breeding with increasing doses of docetaxel.Fluorescence two-dimensional difference gel electrophoresis(DIGE) was used to quantatively screen the differentially expressed proteins between untreated PC-3 cells and apoptotic PC-3 cells,docetaxel-sensitive PC-3 cells and docetaxel-resistant PC-3 cells.The constituents were analyzed by matrix-assisted laser desorption/ionizafion time of flight mass spectrometry(MALDI-TOF-MS) and the expression of HSP60 was detected.Results The expression of HSP60 was up-regulated in docetaxel-induced apoptotic PC-3 cells,and down-regulated in docetaxel-resistant PC-3 cells.Conclusion The expression level of HSP60 is high in the apoptotic PC-3 cells and low in the docetaxel-resistant PC-3 cells.It suggests that HSP60 might play an important role in apoptosis and docetaxel-resistance of PC-3 cells.
