摘要
目的:观察补肾活血中药天鼓冲剂对肾虚感音神经性耳聋豚鼠听力的干预效果并探讨其作用机制。方法:采用肾虚耳聋模型,测定各组动物模型的ABR阈值及潜伏期,光镜下观察毛细胞显微结构及血管纹充盈情况,透射电镜下观察耳蜗听毛细胞酶的活性。结果:与模型组比较,中药治疗组豚鼠ABR阈值及潜伏期明显降低或缩短,毛细胞形态、血管充盈状态及酶活性明显改善。结论:天鼓冲剂对早期肾虚感音神经性耳聋豚鼠有一定的治疗作用,其治疗作用可能与改善内耳微循环、提高内耳Ca2+-ATP酶、Na+-K+-ATP酶活性密切相关。
Objective:To observe the intervene effect of tonifying kidney and promoting blood circulation Chinese medicine(tiangu infusion) on guinea pigs with sensorineural deafness caused by kidney deficiency and explore its mechanism. Methods:Models with deafness were made by injecting kanamycin, than injecting prednisolone acetate to make kidney-deficiency deafness models, guinea pigs were divided into three groups, normal, model and Chinese medicine groups, Chinese medicine group was given tiangu infusion for 30 days. ABR threshold and latency of wave interval of each guinea pig were determined, hairy cell microstructure and stria vascularis filling condition were observed by light microscope, cochlea hairy cell enzyme activity was observed by transmission electron microscope. Results:To compare with normal group, ABR threshold of model guinea pigs was obviously increased, latency of wave interval was significantly prolonged, ABR threshold and latency of wave interval were obviously improved in tiangu infusion group, there were significant differences between normal and model, model and tiangu infusion groups. The morphology of hairy cell, vascular stria filling condition and cochlea hairy cell enzyme activity got better obviously than those in the models. Conclusion:Tiangu infusion can effectively treat sensorineural deafness caused by kidney deficiency at early stage, its mechanism was related to improve inner ear microcirculation, raise inner ear Ca+-ATP ase and Na+-K+-ATP ase activities.
