摘要
目的探讨抱石莲中绿原酸的最佳提取工艺。方法建立高效液相法测定绿原酸含量,采用超声波提取的方法对药材进行提取,以抱石莲中绿原酸得量为参考指标,采用单因素实验、正交实验和方差分析对提取工艺进行考察。结果绿原酸含量检测回归方程为Y=3.72×106X-0.72×106,r=0.999 7(7.8~390.0μg·mL-1)。最佳提取工艺条件为:pH=4.0,乙醇浓度70%,超声时间30 min,料液比1:12,超声提取2次,按此条件进行提取,绿原酸得率为1.67%(n=3)。结论利用单因素考察及正交试验的方法优化绿原酸的提取工艺,为抱石莲中绿原酸的工业化提取和生产奠定了基础。
Objective To investigate the optimal extraction process of chlorogenic acid(CGA) from Lepidogrammitis drymoglossoides(Baker) Ching.Methods HPLC was used to determine the quantity of CGA.Ultrasound extraction was used.Single factor experiment,orthogonal experiments and analysis of variance were all taken to optimize the extraction process,with CGA content as the index.Results The regression curve for CGA analysis was Y=3.72×106X-0.72×106,r=0.999 7(7.8-390.0 μg·mL-1).The optimal conditions for extracting were as follows: pH=4.0,70% ethanol concentration,30 min ultrasound extraction,1 : 12 solid-liquid ratio,twice.The yield of CGA was 1.67%(n=3).Conclusion Single factor experiment and orthogonal test can help optimize the extraction process of chlorogenic acid,which lays the foundation for industrial extraction and production of CGA from Lepidogrammitis drymoglossoides.
