摘要
目的:研究含饱和氢气的HC-A肾保存液对大鼠肾脏冷缺血再灌注损伤的影响。方法:健康雄性Wistar大鼠24只随机分为3组(n=8)。H1组大鼠仅接受右肾切除;H2组大鼠为冷缺血再灌注模型,以腹主动脉插管和下腔静脉缺口作为灌注和流出通路,用4℃普通HC-A肾保存液对左肾行原位冷灌注,灌注完成后阻断左肾动、静脉,缝合腹主动脉和下腔静脉缺口。阻断45min后移除血管夹,冲洗腹腔后关腹;H3组仅灌注液换用自制的4℃含饱和氢气的HC-A肾保存液,其它处理同H2组。3组大鼠均于24 h后再次手术,取血标本,测定BUN和Cr浓度,评估肾功能。切取左肾,检测肾组织中MDA的水平和SOD、caspase-3的活性,并在光镜下观察肾组织形态学的改变。结果:与H2组相比,H3组BUN、Cr、MDA的水平和caspase-3活性降低(P<0.05),SOD活性升高(P<0.05)。光镜下,H3组肾组织病理损伤较H2组减轻。结论:含饱和氢气的肾保存液能减轻大鼠肾脏冷缺血再灌注损伤。
Objective: To investigate the effects of hydrogen-rich HC-A kidney preservation solution on kidney cohl ischernia/reperfnsion injury in rats. Methods: Twenty-four healthy male Wistar rats were randomly divided into 3 groups (n=8). In control group( group H1), only right kidney was removed.Clod ischemia/reperfusion was performed in common HC-A kidney preservation solution group(group H2), a pipe was put in abdominal aorta after the aorta was occluded proximal and distal to the left renal artery to perfuse the left kidney by 4 ℃ common HC-A kidney preservation solution,and a drainage vent was made in inferior vena cava after its being occluded near the junction with the left renal vein. The renal pedicle of left kidney was interdicted after cold perfusion for 45 rain. During that time, the involved vessels were repaired and the other clamps were released.When cold ischemia was completed,the interdiction in the left renal pedicle was removed. Close the abdominal cavity after washing. The perfusion solution of group H3 was hydrogen-rich HC-A kidney presmwation solution, it was the only differenee with group H2. After 24 hours, serum of all rats in three groups were obtained by centrifung ing the venous blood to detect the level of BUN and Cr. Left kidneys were removed to detect the level of MDA and the activity of SOl) and caspase-3 in renal tissue.The pathological ehanges of kidney was evaluated by light microscopy. Results: Compared with group H2, the level of BUN, Cr, MDA and the activity of caspase-3 decreased in group H3 (P〈0.05), while the activity of SOl) rised ( P〈0.05 ). The injury. of group H2 and H3 was obvious,while group H3 was more slight. Conclusion: Hydrogen-rich HC-A kidney preservation solution can attenuate kidney cold ischemia/reperfusion injury in rats.
