摘要 目的 探索和建立西藏小型猪毛乳头细胞体外分离和培养的方法。方法 分离2月龄西藏小型猪毛乳头细胞进行体外原代培养及传代培养,观察毛乳头细胞的形态和生长状况。结果 西藏小型猪毛乳头细胞体外分离培养后,增生速度快,早期培养的细胞呈凝集性生长,而6代后的细胞则呈非凝集性生长。结论 利用分离酶和Ⅳ型胶原酶消化法所获得的西藏小型猪毛乳头细胞可在体外稳定培养并传代。 Objective To explore and establish a method of isolation and cultivation of dermal papilla cell in vitro. Methods Porcine dermal papilla cells were isolated from 2 month Tibet miniature pig and then were cultured and subcultured in vitro to observe morphology and cell growth. Results Tibet miniature pig dermal papilla cells grew quickly in vitro, the early passage cultured porcine dermal papilla cells exhibited an aggregative behavior, but the sixth passage later cultured porcine dermal papilla cells exhibited non-aggregative behavior. Conclusion The Tibet miniature pig dermal papilla cells digested with dispase and collagenase 1V could grow and proliferate in vitro.
