摘要
目的应用比较蛋白组学研究胃癌腹膜转移形成相关的蛋白质分子标志物。方法以胃癌细胞株(BGC823)和腹膜高转移潜能的胃癌细胞株(SCG7901)为研究对象,采用双向凝胶电泳(2-DE)对其进行总蛋白的分离,两组之间的差异蛋白通过质谱分析和蛋白数据库搜索鉴定,并用Westernblot法对差异蛋白点进行进一步检测验证。结果腹膜高转移潜能的胃癌细胞株(SCG7901)与胃癌细胞株(BGC823)比较,共鉴定出20个有意义的蛋白质,其中11个蛋白仅在SCG7901细胞株中表达明显增高,9个蛋白仅在BGC823细胞株中表达明显增高。对于SCG7901细胞株高表达的叉头蛋白3(Foxp3),应用Westernblot法在蛋白水平上验证了2-DE结果。结论腹膜高转移潜能的胃癌细胞株(SCG7901)与胃癌细胞株(BGC823)的蛋白质表达差异有统计学意义。
Objective To study the key proteins associated with metastasis of gastric cancer, and the potential diagnostic markers and therapeutic targets of this malignancy. Methods A comparative pro- teomic strategy, in a combination of two-dimensional electrophoresis separation and mass spectrometry iden- tification, was adopted to search for proteome alternations in an gastric cancer mother cell line BGC823 and its highly peritoneal metastatic cell subline SCG7901. Results There were 20 significant proteins identified in total. Among the 20 spots, 11 proteins were up-regulated in cell line SCG7901, whereas 9 were up-regu- lated in cell line BGC823. Western blotting confirmed the difference of forkhead box I)3 (Foxp3) on protein level. Conclusion The results suggest that an obviously differential proteomic expression exists between the human gastric cancer mother cell line BGC823 and its highly peritoneal metastatic cell subline SCG7901.
