Metabonomic studies of serum obtained from oral cancer patients using 1H NMR spectroscopy

摘要 目的应用核磁共振代谢组学方法研究口腔癌患者血清代谢组的特征及其变化。方法健康人血清(健康组,10例)和口腔癌患者血清(口腔癌组,10例)样本,采用BRUKER 500 MHz超导核磁共振波谱仪进行检测,运用PCA分析方法,研究口腔癌患者血清代谢组的特征,探讨相关生化过程的改变。结果健康组与口腔癌组患者血清代谢谱有不同的代谢特征。口腔癌患者血清中氨基酸(亮氨酸、异亮氨酸、缬氨酸、酪氨酸、苯丙氨酸、组氨酸),谷氨酸、糖蛋白、甲胺和二甲胺、葡萄糖的含量是升高的,而乳酸和胆碱的含量降低。结论根据血清代谢物的变化可以区分健康人和口腔癌患者。这种基于核磁共振氢谱的代谢组学方法可为口腔癌的诊断提供可靠代谢依据。 Objective To research the metabolite compositions of the serum samples from oral cancer patients by using nuclear magnetic resonance (NMR) based metabonomics. Methods NMR ~H spectra were measured for serum samples from 10 healthy volunteers and 10 oral cancer patients. The spectra data were analyzed by principal component analy- sis (PCA) to reveal the metabonomic profiles of serum from oral cancer patients and the corresponding biochemical changes. Results There were obvious metabonomic differences between healthy volunteers and oral cancer patients which were highlighted by the increased levels in amino acids (leucine, isoleucine, valine, tyrosine, phenylalanine, his- tidine), glutamate,glycoprotein, methylamine, dimethylamine and glucose, together with the decreased levels of lactate and choline in oral cancer patients. Conclusion The results show that the metabonome of serum from oral cancer pa- tients differ markedly from that of the healthy volunteers, which indicates that serum NMR 1H spectroscopy in con- junction with PCA offers an efficient tool for the diagnosis of oral cancer.
