摘要
目的:探讨培土生金法对慢性阻塞性肺疾病(COPD)大鼠肺功能以及线粒体超微结构的影响。方法:选用健康Wistar雄性大鼠120只,随机分为6组:即COPD模型组、空白对照组、黄芪建中汤组(高、中、低剂量组)、金水宝胶囊组,气管内滴入脂多糖(LPS)加烟熏制成COPD模型。COPD模型制备完毕后,黄芪建中汤组(高、中、低剂量组)、金水宝胶囊组每天灌胃1次,共8周。实验结束后检测大鼠肺功能,各组大鼠肺组织进行透射电镜观察。结果:大鼠肺功能检测与模型组比较,黄芪建中汤(高、中、低剂量组)、金水宝胶囊组最大呼气流速(PEF)、第0.3秒用力呼气容积(FEV0.3)明显升高,有显著性差异(P<0.05),黄芪建中汤低剂量组FEV0.3/FVC升高不明显,无显著性差异(P>0.05);与黄芪建中汤中、低剂量组及金水宝胶囊组比较:黄芪建中汤高剂量组FEV0.3/FVC、PEF、FEV0.3明显升高(P<0.05)。透射电镜观察,模型组见肺泡Ⅱ型细胞板层小体有明显排空和空泡化改变,线粒体明显肿胀变性,嵴突破坏甚至消失,黄芪建中汤高剂量组线粒体轻度肿胀、结构完整,嵴突大部分完好。其余治疗组大鼠肺泡Ⅱ型细胞板层小体排空和空泡化改变减轻,但不如高剂量组。结论:培土生金法的治疗作用明显优于金水宝胶囊组,能明显改善慢阻肺大鼠肺功能,修复线粒体的结构,调节COPD能量代谢从而延缓COPD的进程。
Objective :To disscuss rat lung function and ultrastructure of mitochondria by building up the spleen to supplement lung. Methods:Total 120 healthy male Wistar rats were randomly divided into 6 groups : COPD model group,normal group,treatment group with RadixAstragali (RA) group (high,medium and low dose group),Jinshuibao capsule group. COPD models were established by smoke inhalations and intratracheal instillations of lipopolysaecharide. After copying the model of COPD,RA group (high,medium and low dose group),Jinshuibao capsule group garage once daily for 8 weeks. Then we detected lung function of the rats. Lung tissues were observed by transmission electron microscopy. Results:The rats lung function testing,FEV0.3/FVC,PEF,FEV0.3 in the model group were significantly lower than the normal group,compared with the normal group(P〈0.05). The PEF,FEV0.3 were significantly difference in the treatment groups compared with the model group(P〈0.05),and FEV0.3/FVC in the low-dose group was not significantly difference compared with the model group(P〉0.05). Compared with the medium,low dose group and Jinshuibao capsule control group,the FEV0.3/FVC,PEF,FEV 0.3 in the RA high dose group was significantly higher(P〈0.05). Transmission electron microscopy observation,in the model group,the ultra-structural changes of type Ⅱ pneumocyte included vacuolation of lamellar bodies and partial cell desquamation. Mitochondrial swelling obvious degeneration and groups,the mitochondria mild swelling,structural crest breakthrough bad or even disappear. The treatment integrity,cristae most intact. The RA high dose group, similar but less profound pathologic changes were found in others groups. But not as good as the high dose group. Conclusion:The effect of building up the spleen to supplement the lung was significantly superiorer than Jinshuibao capsule group. It could significantly improve lung function in COPD rats. Regulation of COPD energy metabolism and delay the process of COPD.
