HPLC法测定腺毛黑种草子中山柰酚

Determination of kaempferol in seeds of Nigella glandulifera by HPLC

摘要 目的建立腺毛黑种草子中山柰酚的HPLC测定方法。方法采用高效液相色谱法,色谱柱为SymmetryRPC18(150mm×4.6mm,5μm)色谱柱,流动相为甲醇-0.4%磷酸水溶液(70:30),体积流量1.0mL/min,检测波长365nm,进样量10μL。结果山柰酚在0.0224~0.1344μg呈良好的线性关系(r=0.9999),平均回收率为97.59%,RSD值为1.29%(n=9)。结论该方法操作简便、准确、重复性好,可用于腺毛黑种草子中山柰酚的测定。 Objective To establish an HPLC method for determination of kaempferol in the seeds of Nigella glandulifera.Methods HPLC was carried out on Symmetry C18 column (150 mm×4.6mm,5μm) with methanol-0.4% phosphoric acid (70:30) as mobile phase.The detection wavelength was 365nm and injection volume was10 μL at the flow rate of 1.0 mL/min.Results The calibration curve was linear over the range of 0.022 4—0.134 4μg (r=0.999 9) for kaempferol.The average recovery rate was 97.59% with RSD value at 1.29%.Conclusion The established method is rapid,sensitive,and accurate,and could be used for the determination of kaempferol in the seeds of N.glandulifera.
