摘要 目的探讨二十二碳六烯酸(DHA)对人肝癌细胞株Bel-7402侵袭力及基质金属蛋白酶9(MMP-9)表达的影响。方法①采用台盼蓝拒染实验检测不同浓度DHA(0、25、50、100、200μmol/L)作用48 h后对细胞存活率的影响;②用Transwell小室模型和细胞划痕实验检测各组细胞的侵袭和迁移能力的变化;③运用Western blot法和细胞免疫组化法检测MMP-9表达的差异。结果不同浓度的DHA均可抑制细胞活率(P<0.05),其程度呈时间-剂量依赖性;与对照组细胞比较,细胞经不同浓度DHA刺激后其侵袭及迁移能力均有所下降(P<0.05);不同浓度DHA作用肝癌细胞48 h后MMP-9的相对表达量均明显降低(P<0.05),其影响呈剂量相关性。结论 DHA能降低人肝癌细胞株Bel-7402的相对活率,并且在一定程度上降低细胞的侵袭及迁移能力,其作用机制可能与抑制MMP-9的表达相关。 Objective To investigate the effects of docosahexaenoic acid on cell invasive ability and MMP-9 expression of human hepatocellular carcinoma cells Bel-7402. Methods (1) The effects of different concentrations of DHA(0, 25, 50, 100, 200 μmol/L) on cell viability were assessed by trypan blue dye exclusion assay. (2) Cell invasion and migration assays were measured by transwell and wound scratch assay. (3) Immunohistochemisty and Western blot assays was employed to determine the expression of MMP-9. Results Different concentrations of DHA could inhibit cell viability ( P 〈 0.05) , and it was in the time and dose-dependent. Compared with control group, the invasion and migration of HCC cells which were treated with different concentrations of DHA had been decreased (P 〈 0. 05 ). MMP-9 relative expression levels were significantly reduced by different concentrations of DHA after 48 hours (P 〈 0.05 ), and the effects were in dose-dependent. Conclusion DHA may inhibit cell via- bility of the human hepatoma cell line Bel-7402, and to a certain degree reduced the invasion and migration of HCC cells. The mechanism may be related to the inhibition of the expression of MMP-9.
