摘要
目的:分析并探讨绒毛组织中抑癌基因PTEN和抗氧化蛋白PeroxiredoxinⅡ(Prx2)与自然流产的关系。方法:应用RT-PCR法及免疫组织化学SP法检测PTEN和Prx2蛋白及mRNA在30例自然流产患者(实验组)绒毛组织中的表达,并以同期非意愿妊娠要求人工流产的30例正常妊娠患者为对照组。结果:①实验组绒毛组织中PTEN和Prx2蛋白阳性表达率(70.0%,36.7%)明显低于对照组(93.3%,93.4%),差异有统计学意义(P<0.05)。②实验组绒毛组织中PTEN和Prx2 mRNA的表达(0.929±0.100,1.131±0.125)明显低于对照组(0.059±0.010,0.222±0.029),差异有统计学意义(P<0.05)。结论:PTEN和Prx2在自然流产患者绒毛组织中表达明显下降,说明可能与自然流产的发生有一定关系。
Objective: To explore the relation of phosphate and tension homology deleted on chromsome ten (PTEN) and peroxiredoxin Ⅱ (Prx2) to spontaneous abortion. Methods: The expression of PTEN and Prx2 in villi tissue of spontaneous abortion (experimental group, n=30) and unwanted pregnancy (control group, n=30) was detected using immunohistochemistry and RT-PCR. Results: 1)The positive expression of PTEN and Prx2 protein in experimental group (70.0%, 36.7%) was significantly lower than that of the control (93.3%, 93.4%)(P〈0.05). 2)The expression of PTEN and Prx2 mRNA in experimental group (0.059 ± 0.010, 0.222 ± 0.029) was also significantly lower than that of the control (0.929 ± 0.100, 1.131 ± 0.125)(P〈0.05). Conclusion: Abnormal expression of PTEN and Prx2 in the villi tissue may have a certain role in the pathophysiology of spontaneous abortion.
