TRAIL对耐药相关基因SIRT1的作用 被引量:1

摘要 目的观察肿瘤坏死因子相关诱导凋亡配体(TRAIL)在食管癌细胞中的表达及产生的凋亡作用。方法构建真核表达载体pcD-NA3.1(+)-TRAIL利用脂质体瞬时转染人食管癌细胞EC9706,并设立转染空载体组和未转染组作为阴性对照;48 h后用RT-PCR和免疫细胞化学方法分别检测其在食管癌细胞中mRNA和蛋白水平的表达;流式细胞技术检测转染重组质粒24、48 h后对食管癌细胞产生的凋亡效应;用RT-PCR和免疫细胞化学方法检测SIRT1 mRNA和蛋白表达水平。结果重组真核表达载体pcDNA3.1(+)-TRAIL中TRAIL基因能大量表达且对食管癌EC9706细胞产生凋亡效应,并能显著抑制耐药基因SIRT1的mRNA和蛋白水平的表达。结论 TRAIL可抑制食管癌细胞中SIRT1基因表达。
