摘要
目的该研究旨在建立胰岛素抵抗大鼠模型,观察高脂喂养大鼠脂肪细胞因子的表达,研究胰岛素抵抗的发生机制。方法普通级雄性Wistar大鼠24只,称重后随机分为高脂喂养组、IL-6注射组、正常对照组;6周后所有大鼠禁食12h后称重,测FBG、FINS,并按HOMA公式计算胰岛素抵抗指数(HOMA.IR);ELISA法测定血清IL-6、脂联素水平。结果与正常对照组相比,高脂喂养组血清IL-6水平明显升高,脂联素水平显著下降(P〈0.05);IL-6注射组的脂联素水平降低(P〈0.05);相关分析显示,IL-6与HOMA—IR正相关(r=0.75,P〈0.05),与脂联素负相关(r=0.78,P〈0.01)。结论IL-6参与胰岛素抵抗发生机制,可能至少部分是通过减少脂联素的分泌。
Objective To investigate the mechanism of insulin resistance in high-fat diets rats.Method Twenty-four male Wistar rats were randomly divided into 3 groups after weighting:the first group were fed with high-fat diet,the second group were injected with IL-6,the third group were fed with normal diet as control.After six weeks,FBG and FINS were measured and HOMA-IR was assessed by homeostasis mode assessment.The level of adiponectin and interleukin-6 was detected by ELISA.Results The serum level of IL-6 in high fat diet rats was significantly higher and the level of adiponectin was lower compared with the control group(P 〈 0.05). Interestingly, the level of adiponectin decreased after injection of IL-6(P 〈 0.05).Correlation analyze indicated that the level of IL-6 was positively correlated with HOMA-IR(r=0.75,P 〈 0.05),while negatively correlated with adiponectin(r=-0.78,P 〈 0.01).Conclusion IL-6 may contribute to one of the mechanisms of insulin resistance by decreasing adiponectin secretion.
