金莲花中荭草苷和牡荆苷小鼠体内抗氧化作用研究 被引量:8

摘要 目的研究金莲花中黄酮单体荭草苷和牡荆苷对小鼠全血以及组织中抗氧化酶活性、丙二醛含量的影响。通过比较荭草苷和牡荆苷抗氧化作用,筛选出抗衰老作用最佳药物的构效关系。方法昆明种小鼠70只,随机分成正常对照组、荭草苷和牡荆苷各3个剂量组,连续给药12d,对照组给予同体积溶剂,d13进行断头取血和各组织器官、血浆离心得血清,用生理盐水制成10%的组织匀浆液,8 000r·min-1离心得上清液,试剂盒测定小鼠血清以及组织中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-PX)活力及组织中丙二醛(MDA)含量。结果不同浓度的荭草苷和牡荆苷均具有提高小鼠血清及组织SOD、CAT、GSH-Px的活性和降低小鼠组织MDA含量的作用;荭草苷和牡荆苷各剂量组与对照组相比差异具有统计学意义(P<0.05或P<0.01)。荭草苷中、低剂量组在提高小鼠血清及组织SOD、CAT、GSH-Px的活性、降低组织MDA含量上高于牡荆苷同等剂量组,差异具有统计学意义(P<0.05);荭草苷和牡荆苷高剂量组相比,差异无显著性。结论荭草苷和牡荆苷具有较好的抗氧化作用,荭草苷抗氧化活性优于牡荆苷。 Objective To study the effects of antioxidation and Malondialdehyde(MDA)of orientin and vitexin in Trollius chinesis to mice,investigate active components in Trollius chinesis,and expect to discover new type of substances for antioxidation.Methods The mice were divided into blank,high,medium and low dose groups of orientin and vitexin.Every morning mice had intragastric administration,for 12 days.After decapitated on 13th day,blood was selected,and centrifugation was made for serum.Then serum antioxidant enzymes including superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-PX)were detected.And also liver,brain and kidney were cutted in frozen conditions,10% tissue homogenate was made with NS solution,supernatant was got by centrifugation at 8 000 rpm,and levels serum antioxidant enzymes including SOD,CAT,GSH-PX and MDA were detected.Results Orientin and vitexin significantly improved the activity of SOD,CAT and GSH-PX of mouse blood serum,and increased activity of SOD,CAT and GSH-PX of liver,brain and kidney,and reduced activity of MDA.Compare with those of normal group,orientin and vitexin of each dose group showed significant difference(P0.05,P0.01).The activity of medium and low dose group of orientin was better than that of same dose group of vitexin,and that of the same dose group of orientin and vetixin had significant difference(P0.05).The high dose group of orientin and vetixin had no significant difference.Conclusion Orientin and Vitexin of Trollius Chinesis could apparently improve activity of antioxidation in serum and tissue and reduce the amount of MDA.The activity of orientin was better than that of vitexin.
