果蔬汁饮料DNA提取方法的比较研究 被引量:7

Comparison of DNA extraction method for fruit and vegetable juices

摘要 以澄清型果蔬汁为实验材料,用3种DNA提取方法(普通CTAB法、试剂盒法、改良试剂盒法)提取果蔬汁中DNA片段。以紫外分光光度计测量吸光值计算DNA的纯度和得率,以内源rbcL基因为目标基因建立了PCR扩增技术体系。实验结果表明:试剂盒法和改良试剂盒法均适用于果蔬汁饮料DNA的提取,其中改良试剂盒法提取DNA的纯度和产率要高于另外2种方法,该方法为从果蔬汁加工品提取高质量的DNA并进行分子检测提供技术参考。 Three kinds of DNA extraction method(CTAB,DNA extraction kit,modified DNA extraction kit) to extract genomic DNA fragment from clarified type fruit and vegetable juice.The purity and yield of the DNA were determined by A 260 /A 280 respectively.The PCR amplification of partical rbcL gene fragement was set up.The results showed that both the DNA extraction kit and modified DNA extraction kit were suitable for DNA extraction from fruit and vegetable juices,the DNA purity and yield extracted with modified DNA extraction kit is higher than with the other method in this research.The research provides an effective method for DNA extraction from fruit and vegetable juices.
