抗生素克林霉素在改良MRS检测嗜酸乳杆菌的应用研究

摘要 探讨应用克林霉素改良MRS(Clindamycin-MRS)进行嗜酸乳杆菌计数的最佳条件。使用嗜酸乳杆菌4株对不同剂量Clindamycin-MRS进行性能测试;对5份含嗜酸乳杆菌的市售样品进行嗜酸乳杆菌计数;并观察嗜酸乳杆菌在不同培养时间的生长情况。结果表明当克林霉素(Clindamycin)的添加剂量为0.05~0.15μg/100 mL时,嗜酸乳杆菌4株试验菌株在Clindamycin-MRS琼脂中的生长率PR均在0.98~1.23间,生长良好;克林霉素(Clindamycin)的添加剂量大于2.0μg/100 mL时,嗜酸乳杆菌均不生长。5份样品中,克林霉素(Clindamycin)的添加剂量在0.05~0.10μg/100 mL时,嗜酸乳杆菌计数1.0×10~6~4.6×10~9CFU/mL,生长良好;非目标乳酸菌完全受抑制。嗜酸乳杆培养厌氧培养48±2 h,菌落细少;培养(72±2)h,形成易于计数的菌落。结论为用Clindamycin-MRS琼脂进行嗜酸乳杆计数时,克林霉素(Clindamycin)的添加剂量可选0.05~0.10μg/100 mL,培养时间选择(72±2)h。 To investigate the optimum culture condition of enumeration of Lactobacillus acidophilus in Clindamycin- MRS Medium. Use the Lactobacillus acidophilus 4 strains to the performance test for different doses of Clindamycin- MRS. The 5 samples had Lactobacillus acidophilus. The growth features ofLactobacillus acidophilus were observed in different incubation times. The results showed the productivity ofLactobacillus acidophilus were all among 0.98±1.23 in Clindamycin-MRS media with 0.05-0.15μg/100 mL of Clindamycin and well growth. The Lactobacillus acidophilus failed to growth in Clindamycin-MRS media with Clindamycin ≥2.0 μg/100 mL. Lactobacillus acidophilus counted of 1.0× 106~4.6×109 CFU/mL in Clindamycin-MRS media with 0.05-0.10 μg/100 mL of Clindamycin and well growth. Non-target strains could be totally inhibited. Lactobacillus acidophilus could only form the pinpoint colonies after (48±2) h incubation, but all the colonies were moderate sizes after 72±2 h incubation. The conclusion suggested that the optimum Clindamycin in does was 0.05~0.10μg/100 mL and the optimum incubation time was 72±2 h using Clindamycin-MRS media for enumeration of Lactobacillus acidophilus in food.
